The study was a double-blind, placebo-controlled, parallel group trial. Patients, who had a diagnosis of schizophrenia, bipolar I disorder (BID), or major depressive disorder (MDD), were recruited to participate in the study. Eligible patients were randomized to receive 300 mg quetiapine or placebo (N=3437) for 3 weeks. A total of 972 patients (0.5% of the study population) were assigned to quetiapine (mean age, 64 years; 95% CI, 69.3-71.7) or placebo (n=3437) for a total of 4 weeks, and 663 patients (0.5% of the study population) were assigned to receive a 7-day course of treatment. Treatment was continued for a total of 7 days. The study was stopped for an additional 8 days after completion of the study. Patients with severe depression or bipolar disorder, who were randomized to quetiapine or placebo, were also included in the study. The study was approved by the institutional review board of the University of California, San Francisco (UCSF, San Francisco CA).
Results
The total number of patients assigned to quetiapine for 3 weeks was 972. The incidence of adverse events was statistically significantly higher with quetiapine (1.5% versus 1.8%;P=.039) as compared to placebo (4.9% versus 7.9%;<.0001).
